030_ Phenotypic variability of cognitive, psychosocial functioning, and symptomatic data in the PsyCourse cohort.
Research Question and Aims
A focus of this proposal is the systematic investigation of phenotypic variability in cognition, psychopathology, and psychosocial impairment in the PsyCourse cohort. This issue is raised in the KFO241 grant application, WP5. We will use the results of this proposal in the KFO241/PsyCourse grant report due in summer, but depending on the results aim at an independent journal publication. The main question is how constant phenotypes assessed in controls stay over the 18 months assessment period with respect to both variability and trend. Furthermore, we would like to compare this putative longitudinal constant behavior and/or variability to that of cases. For this purpose, we will use where appropriate linear mixed models, but also methods independent of such distributional assumptions like the rang-based longitudinal nonparametric test (LNTP, PMID: 20568282), previously extended by our group to the context of genetic epidemiology, and successfully applied to analyze other cohorts (e.g. Psychosis, PMID: 26475575).
Analytic Plan
First we have selected several phenotypes (see list below) in order to focus this investigation. We will start with descriptive statistics regarding variability and course and missingness of assessments. We will then use LNPT and linear mixed models to assess time effects (analyses of control participants) and time*diagnosis (comparison of cases and controls). A special emphasis will be given to handling of age classes and varying time-points in the LNPT (KFO241, WP5, aims 2 and 4). Correction for multiple testing will not be applied to between phenotypes for those analyses using only controls, as it is important to identify potential non-constant and variable patterns in controls that might create unexpected noise for comparisons between cases and controls. Thus this part has a more explorative nature (consider also the limited sample size of roughly 300 controls). Comparisons with cases will be carried out with appropriate corrections.
